The Algerian side finished as runners-up in Group B behind DR Congolese giants TP Mazembe having accumulated eight points from six games.
As a result, Setif booked their place in the last eight of Africa's most prestigious club competition.
They will look to 29-year-old Algerian striker El Habib Bouguelmouna when they lock horns with Wydad at home.
Meanwhile, Wydad dominated Group C which they won ahead of runners-up Horoya AC of Guinea having collected 12 points from six games.
The dominance in the group stage earned the Moroccan giants a place in the quarter-finals.
Wydad's dangerman is 26-year-old Moroccan winger Mohamed Ounajem, who will be looking to inspire his side to victory in Algeria.
The Wydad of Resistance are looking to defend their Champions League title having won it last year.
Gabonese match referee Eric Otogo-Castane will officiate the game between Setif and Wydad.
